一抗孵育(Primary antibody incubation)

參考一抗的說(shuō)明書(shū)，按照適當(dāng)比例用Western一抗稀釋液稀釋一抗。
用微型臺(tái)式真空泵吸盡封閉液，立即加入稀釋好的一抗，室溫或4℃在側(cè)擺搖床上緩慢搖動(dòng)孵育一小時(shí)。如果一抗孵育一小時(shí)效果不佳，可以在4℃緩慢搖動(dòng)孵育過(guò)夜?；厥找豢?。加入Western洗滌液，在側(cè)擺搖床上緩慢搖動(dòng)洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長(zhǎng)洗滌時(shí)間并增加洗滌次數(shù)。

二抗孵育(Secondary antibody inucubation)

參考二抗的說(shuō)明書(shū)，按照適當(dāng)比例用Western二抗稀釋液稀釋辣根過(guò)氧化物酶(HRP)標(biāo)記的二抗。
用微型臺(tái)式真空泵吸盡洗滌液，立即加入稀釋好的二抗，室溫或4℃在側(cè)擺搖床上緩慢搖動(dòng)孵育一小時(shí)?；厥斩梗尤隬estern洗滌液，在側(cè)擺搖床上緩慢搖動(dòng)洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長(zhǎng)洗滌時(shí)間并增加洗滌次數(shù)。

英文介紹：GSTZ1 is a member of the glutathione S transferase (GSTs) super family, encoding multifunctional enzymes important in the detoxification of electrophilic molecules, including carcinogens, mutagens, and several therapeutic drugs, by conjugation with glutathione. This enzyme also plays a significant role in the catabolism of phenylalanine and tyrosine. Several transcript variants of this gene encode multiple protein isoforms. GSTZ1 shows minimal glutathione-conjugating activity with ethacrynic acid and 7-chloro-4-nitrobenz-2-oxa-1,3-diazole and maleylacetoacetate isomerase activity. It has low glutathione peroxidase activity with T butyl and cumene hydroperoxides and is able to catalyze the glutathione dependent oxygenation of dichloroacetic acid to glyoxylic acid. Highest expression in liver followed by kidney, skeletal muscle and brain. Also expressed in melanocytes, synovium, placenta, breast and fetal liver and heart.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
