Gibco CTS AAV-MAX Lysis Buffer is a ready-to-use cell lysis reagent for the extraction of adeno-associated virus (AAV) particles from producer HEK293 cells. The cell lysis is detergent-based, eliminating the need for mechanical cell disruption, providing a milder and easier alternative when isolating AAV particles from cell cultures. The buffer is supplied as a 10X solution that can be directly added to HEK293 AAV production cultures to induce cell lysis.
Note: the formulation of this product differs from the formulation of our research-use-only AAV-MAX Lysis Buffer (Cat. No. A50520). This formulation reduces sample turbidity and precipitation formation post-clarification, including when samples are stored between clarification and subsequent downstream workflow steps.
Features of CTS AAV-MAX Lysis Buffer include:
• Polysorbate 20-based formulation that does not contain Triton X-100
• Formulated with 20 mM MgCl2 for convenience
• Manufactured in conformity with cGMP for medical devices (21 CFR Part 820) and follows USP and European Pharmacopoeia (Ph. Eur.) 5.2.12 recommendations. Drug Master File Letter of Authorization and Regulatory Support File available upon request.
The CTS AAV-MAX Helper-Free AAV Production System is a scalable, high-titer platform for suspension AAV production with research-grade and CTS versions to enable seamless transition from research to commercial manufacturing. Other components of the CTS AAV-MAX system include CTS Viral Production Cells 2.0, CTS AAV-MAX Transfection Kit, CTS Viral-Plex Complexation Buffer, and Viral Production Medium, AGT.
