KLB蛋白抗體,Anti-KLB抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

KLB蛋白抗體,Anti-KLB抗體石蠟切片的免疫組化SP法參考步驟：

1.60℃烤片2小時
2.脫蠟。二甲苯脫蠟15分鐘，三次
3.水化。依次經(jīng)過無水乙醇、95%、90%、80%、70%，各5分鐘，蒸餾水（或自來水）5分鐘
4.PBS洗5分鐘，三次
5.抗原修復(fù)。枸櫞酸緩沖液（約92-95℃）煮沸（沸水?。┬迯?fù)15分鐘，自然涼至室溫。PBS洗5分鐘，三次
6.3%雙氧水消化內(nèi)源性的過氧化物酶，20分鐘，PBS洗5分鐘，三次
7.正常山羊血清封閉，37℃20分鐘
8.一抗孵育，4℃過ye.
9.PBS洗5分鐘，三次
10.二抗孵育，37℃20分鐘（二抗為*標(biāo)記的羊抗兔IgG），PBS洗5分鐘，三次
11.三抗孵育，37℃20分鐘（三抗為辣根酶標(biāo)記的鏈霉親和素），PBS洗5分鐘，三次
12.DAB顯色，鏡下觀察結(jié)果，適時終止
13.蘇木素復(fù)染5分鐘，自來水沖洗片刻，1%的鹽酸酒精（75%）溶液（請自行配置，配制比例為：100ml 75%酒精+1ml濃鹽酸）分化30秒，自來水沖洗5分鐘藍化
14.脫水。依次經(jīng)過70%、80%、90%、95%、無水乙醇，各5分鐘，二甲苯15分鐘，三次
15.封片，中性樹膠封片

KLB蛋白抗體背景介紹：BetaKlotho is a 1044 amino acid single-pass type III membrane protein that plays a key role in bile acid and cholesterol metabolism by suppressing transcription of CYP7A1 (cholesterol 7-alpha-hydroxylase), the rate-limiting enzyme in bile acid synthesis. Homozygous negative betaKlotho mice showed dramatically elevated bile acid synthesis and secretion, as well as a strong upregulation of CYP7A1 and CYP8B1 and resistance to gallstone formation. FGF19 and FGF21 require direct interaction with betaKlotho for activation, intracellular signaling and gene expression modulation. Both Klotho and betaKlotho consist of two internal repeats similar to family 1 glycosidases. BetaKlotho contains two glycosyl hydrolase 1 regions, however since these regions lack essential glutamic acid residues at specific and crucial locations, the domains appear to be inactive.

方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫。
方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，?；?min，中火5min，自然冷卻至室溫。
方法3：高壓修復(fù)，修復(fù)液加入高壓鍋加熱至沸騰，放入玻片，封蓋加壓持續(xù)加熱至噴氣時開始計時修復(fù)2min，自然冷卻至室溫。
方法4：使用*修復(fù)，將*修復(fù)液加入到組織上，37℃，消化30min
