背景介紹:Drebrins (developmentally regulated brain proteins) are cytoplasmic proteins that bind F-actin in the brain and are involved in cell migration, extension of neuronal processes and plasticity of dendrites. HIP-55 (HPK1-interacting protein of 55 kDa), also known as ABP1, SH3P7 or DBNL (drebrin-like), is a 430 amino acid cytoplasmic protein that belongs to the ABP1 family. HIP-55 binds to F-actin but is not involved in actin polymerization, capping or bundling. In addition to containing an ADF-H domain, HIP-55 also consists of a SH3 domain, which mediates interaction with SHANK2, SHANK3 and PRAM-1. HIP-55 acts as an actin-binding adapter protein and as a common effector of antigen receptor-signaling pathways in leukocytes. As a key component of the immunological synapse, HIP-55 regulates T-cell activation by bridging TCRs and the actin cytoskeleton to gene activation and endocytic processes. HIP-55 is degraded by caspases during apoptosis.

The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

1.抗原進(jìn)入機(jī)體;
2.抗原刺激 B淋巴細(xì)胞,使之分化增殖成為抗體分泌漿細(xì)胞和記憶細(xì)胞(初次免疫應(yīng)答);
3.抗體主要由漿細(xì)胞分泌 ;
4.記憶細(xì)胞再次受到特異性抗原刺激后會(huì)被激活而成為分泌抗體的漿細(xì)胞(再次免疫應(yīng)答);
5.記憶B細(xì)胞是長(zhǎng)命細(xì)胞,可以存活數(shù)月甚至機(jī)體終生 。
初次體液免疫是指宿主*次接觸外來(lái)TD抗原后血清中逐漸出現(xiàn)抗原特異性抗體的過(guò)程。初次體液免疫應(yīng)答的延遲相(從抗原免疫到抗體水平達(dá)到高峰的時(shí)間)為6-10天,起初幾天血清中的抗原特異性抗體主要為親和力較低的IgM,后期以IgG為主。初次應(yīng)答過(guò)程的血清中抗原特異性抗體的高峰濃度較低且維持時(shí)間較短。再次體液免疫應(yīng)答是指曾被某抗原免疫過(guò)的宿主再次接觸該抗原時(shí)血清中迅速出現(xiàn)抗原特異性抗體的過(guò)程,其延遲相僅為4-5天,血清抗體以IgG為主,其高峰濃度較高且維持時(shí)間較長(zhǎng)。再次應(yīng)答所產(chǎn)生的抗體還有親和力成熟現(xiàn)象,即該抗體與抗原的親和力明顯高于初次應(yīng)答產(chǎn)生的抗體。

免疫組化操作步驟:
(一)、儀器設(shè)備
1、 18cm不銹鋼高壓鍋或電爐或用微波爐
2、 水浴鍋
(二)、試 劑
1、 PBS緩沖液(pH7.2―7.4)
2、 0.01mol/L檸檬酸鈉緩沖液(CB,pH6.0,1000ml)
3、 3%甲醇―H2O2溶液:用30% H2O2 和80%甲醇溶液配制
4、 風(fēng)裱劑:
a. 甘油和0.5mmol/L碳酸鹽緩沖液(pH9.0–9.5)等量混合
b. 油和TBS(PBS)配制
5、 TBS/PBS pH9.0–9.5,適用于熒光纖維鏡標(biāo)本;pH7.0-7.4適合光學(xué)纖維標(biāo)本
(三)、操作流程
1、 脫蠟和水化:脫蠟前應(yīng)將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘
a. 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘
b. 無(wú)水乙醇中浸泡五分鐘
c. 95%乙醇中浸泡五分鐘
d. 75%乙醇中浸泡五分鐘
2、 抗原修復(fù):用于福爾馬林固定的石蠟包埋組織芯片
